The answer to your first question is yes.
Note: that it returns 3 volumes.
1. Total volume
2. a positive volume (everything ABOVE the z=0 plane)
3. a negative volume (everything BELOW the z=0 plane)
Qustion #2:
If you create a reference surface (where z=16 for example) use the calculator to subtract the reference from your original and compute the volume of the result.
